Chapter 1: Understanding Microtomes
A microtome is a precision instrument that cuts extremely thin slices of biological or materials specimens for examination under a microscope. The sections it produces are typically between 1 and 50 micrometers thick, with routine histology work commonly targeting 3 to 10 micrometers. For context, a human red blood cell is roughly 7 to 8 micrometers in diameter, so a standard paraffin section is around the thickness of a single cell. This level of precision is unattainable with a scalpel or razor blade in freehand use; the microtome provides a rigid, controlled cutting action so that every section is consistent in thickness across its entire area.
The name comes from the Greek words micros (small) and tome (to cut), but the concept is far older than the word. Early pioneers like Antonie van Leeuwenhoek cut specimens by hand, often producing sections irregular enough to distort their observations. In the late 18th and 19th centuries, instrument makers developed mechanical devices that could hold a specimen steady and advance it a fixed distance against a blade. These first microtomes, built of brass and steel with screw-driven feeds, established the core design that persists today: a block holder, a knife holder, and a mechanism that advances the block a precisely controlled distance after each cut.
